Description: Senicapoc, formerly known as ICA-17043, is a highly potent Gardos Channel (Ca2+-activated K+ channel; KCa3.1) inhibitor/blocker with an IC50 of 11 nM, and with the potential for the treatment of a subset of Hereditary Xerocytosis caused by mutations in the Gardos channel.

ProtocolIn VitroICA-17043 shows Gardos channel that blocks RBCs in mice (C57 black), IC50 is 50±6 nM. ICA-17043 Prevents an increase in cellular hemoglobin concentration in human erythrocytes in a concentration-dependent manner
In VivoICA-17043 (10 mg/kg, oral) Gardos channel activity measured on days 11 and 21 after administration was significantly reduced and correlated with a corresponding increase in erythrocyte K content, while there was no change in Na content. ICA-17043 (10 mg/kg twice daily) induces a significant increase in Hct after 11 days of administration in mice with SAD++
These protocols are for reference only. InvivoChem does not independently validate these methods.

Method for preparing DMSO master liquid: mg drug pre-dissolved in µL DMSO(Master liquid concentration mg/mL) ,Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug.
Method for preparing in vivo formulation: Take µL DMSO master liquid, next add µL PEG300, mix and clarify, next add µL Tween 80,mix and clarify, next add µL ddH2O,mix and clarify.
Note:
  • (1) Please be sure that the solution is clear before the addition of next solvent. Dissolution methods like vortex, ultrasound or warming and heat may be used to aid dissolving.
  • (2) Be sure to add the solvent(s) in order.
