ProtocolIn Vitro Nigericin is more toxic to S18 cells than S26 cells, IC50 They were 2.03±0.55 μM and 4.77±2.35 μM, respectively. Nigericin can selectively kill cancer stem cells in NPCs in vitro. Nigericin significantly reduces the migration capacity of S18 and HONE-1 cells
In VivoNgericin (4 mg/kg, ip) significantly reduced tumor growth and worked synergistically with the chemotherapy agent DDP, as indicated by tumor volume. Nigericin significantly reduces BMI-1 in the body. Overexpression of Bmi-1 partially restored the CSC content and metastasis capacity of NPC cells under Nigerian mycin treatment. The downregulation of BMI-1 may be related to the inhibitory effect of nigmycin on nasopharyngeal carcinoma stem cells
These protocols are for reference only. InvivoChem does not independently validate these methods.

Method for preparing DMSO master liquid: mg drug pre-dissolved in µL DMSO(Master liquid concentration mg/mL) ,Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug.
Method for preparing in vivo formulation: Take µL DMSO master liquid, next add µL PEG300, mix and clarify, next add µL Tween 80,mix and clarify, next add µL ddH2O,mix and clarify.
Note:
  • (1) Please be sure that the solution is clear before the addition of next solvent. Dissolution methods like vortex, ultrasound or warming and heat may be used to aid dissolving.
  • (2) Be sure to add the solvent(s) in order.
