Description: MC-Val-Cit-PAB is a novel cathepsin cleavable ADC linker that is utilized for the synthesis of antibody-drug conjugate (ADC) such as some of the FDA approved ADC drugs including brentuximab vedotin. The mechanism is like this: the Val-Cit will specifically be cleaved by catepsin B as this enzyme is only present in the lysosome, the ADC warhead/payload (the drug) will be released in the cell. The Azido group will react with DBCO, BCN or other Alkyne groups through click chemistry. The hydrophilic PEG spacer increases solubility in aqueous media.
