Description: Digoxin (12β-Hydroxydigitoxin; Dixina; HSDB-214; Novodigal; Lanoxin PG; SK-Digoxin; Cardiogoxin) is a sodium-potassium pump inhibitor and a cardiotonic glycoside that has been approved for use in the treatment of various heart conditions such as arrhythmia and heart failure.

ProtocolIn VitroDigoxin is a sodium-potassium ATPase inhibitor with a negative Z-score of -26.67, indicating that sodium-potassium ATPase plays a role in CHIKV infection. Digoxin-treated U-2 OS cells resulted in a dose-dependent reduction in CHIKV infection relative to DMSO-treated cells, with half the maximum effective concentration (EC).50) is 48.8 nM. Digoxin treatment similarly reduced CHIKV infection in primary human synovial fibroblasts (HSF) and Vero African green monkey kidney cells, EC50 43.9 nM and 67.3 nM, respectively. Digoxin treatment significantly reduces CHIKV infection in these cell types, EC in ST2 cells50 EC in 16.2 μM, C2C12 cells50 is 23.2 μM, which is the EC50 330 and 475 times Digoxin in U-2 OS cells.
