ProtocolIn VitroLiquiritigenin is dose-response activation of ERE tk-Luc production in U2OS cells transfected with ERβ instead of ERα. Liquiritigenin produces dose-dependent activation of ERβ rather than ERα and time-dependent increases in CECR6, NKG2E, and NKD. The ERβ selectivity of Liquiritigenin is due to the selective recruitment of the co-activator steroid receptor co-activator 2 to the target gene. Liquiritigenin exhibits similar binding affinity for ERα and ERβ and results in SRC-2 selectively recruiting target genes into ERβ cells
In VivoIn mouse xenograft models, Liquiritigenin did not stimulate uterine size or tumorigenesis in MCF-7 breast cancer cells
