ProtocolIn Vitro(-)-(S)-Equol inhibits the growth of LnCaP, DU145, and PC3 human prostate cancer cell lines. (-)-(S)-Equol induces apoptosis in PC1 cells by downregulating cyclin B1 and CDK21 and upregulating CDK inhibitors (p27 and p3) and upregulating Fas ligands (FasL), resulting in cell cycle arrest in G2/M phase and pro-apoptotic Bim expression. (-)-(S)-Equol increases the expression of FOXO3a, decreases the expression of p-FOXO3a and enhances the nuclear stability of FOXO3a. (-)-(S)-Equol also reduces the expression of MDM2, which is used as E2 ubiquitin ligase for p-FOXO3a, thereby preventing p-FOXO3a degradation by the proteasome
In VivoAt day 33, (-)-(S)-Equol inhibited tumor growth by 43.2% and 28.4% compared to controls, indicating no significant toxicity of the compound
