ProtocolIn VitroSTF-62247-treated cells accumulated intracytoplasmic vacuoles characteristic of cells undergoing autophagy. Moreover, these vacuoles are larger in VHL-deficient RCC4 and SN12C-VHL shRNA cells than in wild-type VHL cells. In RCC4, RCC4/VHL, SN12C, SN12C-VHL shRNA cells, STF-62247 (0-30 μM) is selectively toxic to VHL-deficient cells compared to their VHL wild-type counterparts.
In VivoSTF-62247 (2.7-8 mg/kg; intraperitoneal injection; daily; for 9 days) treatment significantly reduces tumor growth of VHL-deficient cells.
These protocols are for reference only. InvivoChem does not independently validate these methods.

Method for preparing in vivo formulation: Take µL DMSO master liquid, next add µL PEG300, mix and clarify, next add µL Tween 80,mix and clarify, next add µL ddH2O,mix and clarify.
Note:
  • (1) Please be sure that the solution is clear before the addition of next solvent. Dissolution methods like vortex, ultrasound or warming and heat may be used to aid dissolving.
  • (2) Be sure to add the solvent(s) in order.
