ProtocolIn Vitro2-Methoxyestradiol (2-ME) (5-100 μM) inhibits the assembly of purified tubulin in a concentration-dependent manner, with a maximum (200%) inhibition at 2 μM of 2-Methoxyestradiol (2ME60). In live interphase MCF7 cells, mitotic arrest ICs50 (1.2 μM), 2-Methoxyestradiol significantly inhibits mean microtubule growth rate, duration and length, and overall dynamics, consistent with its role in vitro, and without any observable microtubule depolymerization. 2-Methoxyestradiol induces G in many cell types that divide actively2-M arrest and apoptosis while protecting quiescent cells. 2-Methoxyestradiol binds to tubulin at or near colchicine sites, inhibits microtubule assembly, and high concentrations have been shown to depolymerize microtubules in cells
