Description: Tautomycin is naturally occurring antifungal antibiotic found in shellfish and is produced by the bacterium Streptomyces spiroverticillatus. It is a polyketide-based structure characterized by a three hydroxyl groups, two ketones, a dialkylmaleic anhydride, an ester linkage (connecting anhydride unit to polyketide chain), a spiroketal and one methyl ether among others. Tautomycin is an antifungal antibiotic that is a potent and specific inhibitor of protein phosphatases 1 and 2A and induces contraction of smooth muscle under Ca2+-free conditions, with Kiapp values of 0.16 nM and 0.4 nM for PP1 and PP2A, respectively.

ProtocolIn VitroMTC cell growth is inhibited in a dose-dependent manner by Tautomycin (0-1000 nM, 48 hours)[2]. NE markers ASCL1 and CgA in human MTC cells are decreased in a dose-dependent manner by Tautomycin (0-1000 nM, 48 hours)[2]. Cell Proliferation Assay[2] Cell Line: Human MTC cells Concentration: 0-1000 nM Incubation Time: 48 hours Result: Inhibited MTC cell growth in a dose-dependent manner. Western Blot Analysis[2] Cell Line: Human MTC cells Concentration: 0-1000 nM Incubation Time: 48 hours Result: Led to a dose-dependent decrease in the NE markers ASCL1 and CgA.

Method for preparing DMSO master liquid: mg drug pre-dissolved in µL DMSO(Master liquid concentration mg/mL) ,Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug.
Method for preparing in vivo formulation: Take µL DMSO master liquid, next add µL PEG300, mix and clarify, next add µL Tween 80,mix and clarify, next add µL ddH2O,mix and clarify.
Note:
  • (1) Please be sure that the solution is clear before the addition of next solvent. Dissolution methods like vortex, ultrasound or warming and heat may be used to aid dissolving.
  • (2) Be sure to add the solvent(s) in order.
