Description: (S,R)-GSK321 is a potent and specific inhibitor of mutant IDH1. The IC50s for inhibiting R132G, R132C, R132H and WT IDH1 are 2.9, 3.8, 4.6 and 46 nM respectively. The selectivity is 100 times higher than that of IDH2. . (S,R)-GSK321 induces a decrease in intracellular 2-HG, fails to block myeloid differentiation, and induces granulocyte differentiation at the level of leukemic blasts and more immature stem cells. (S,R)-GSK321 may be utilized in study/research of acute myeloid leukemia (AML) and other cancers.
