ProtocolIn VitroRiluzole is an anticonvulsant that belongs to the family of use-dependent Na-channel blockers that also inhibit GABA uptake, IC+50 is 43 μM. At 20 μM, Riluzole only slightly suppresses peak self-touching IPSC, but reliably lengthens IPSC. Riluzole was also found to cause intense, concentration-dependent, reversible enhancement of the 2 μM GABA response. At higher concentrations of Riluzole, especially 300 μM, GABA currents exhibit significant desensitization upon prolonged co-exposure to 2 μM GABA and Riluzole. Riluzole enhances the EC of GABA reactions50 Approximately 60 μM

Method for preparing DMSO master liquid: mg drug pre-dissolved in µL DMSO(Master liquid concentration mg/mL) ,Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug.
Method for preparing in vivo formulation: Take µL DMSO master liquid, next add µL PEG300, mix and clarify, next add µL Tween 80,mix and clarify, next add µL ddH2O,mix and clarify.
Note:
  • (1) Please be sure that the solution is clear before the addition of next solvent. Dissolution methods like vortex, ultrasound or warming and heat may be used to aid dissolving.
  • (2) Be sure to add the solvent(s) in order.
