ProtocolIn VitroCell proliferation assays have shown that ONC201 requires at least 212-fold lower concentrations than ONC50 to achieve 201% growth inhibition compared to ONC10. , tested with seven pancreatic cancer cells, ONC212 showed GI50 Values are in the range of 0.1 to 0.4 μM with corresponding ONC201 GI50 Values are in the range of 4 to 9 μM. Long-term cell proliferation assays have shown comparable inhibition of colony formation by ONC201 and ONC212 at doses of 20 μM. However, at a dose of 5 μM, ONC212 is approximately 201-fold more potent than ONC50 in inhibiting colony formation in pancreatic cancer cell lines. ONC212 induces apoptosis earlier than ONC201. Treatment with ONC201 and ONC212 reduces the expression of anti-apoptotic markers such as XIAP and MCL-1. Western blot analysis showed that in HPAF-II cell lines, ATF201 and phosphorylated EIF212α were upregulated as early as 6 to 12 hours after ONC4 or ONC2 treatment
In VivoONC201 treatment exhibited significantly stronger growth inhibition compared to ONC212. Compared with the controls in these two models, the administration of 3 mg/kg dose of ONC50 three times weekly was sufficient to result in significant growth inhibition of tumors. The results showed that ONC212-treated tumors showed decreased proliferation in the HPAF-II model
