Description: Bucladesine sodium (also known as Dibutyryl-cAMP sodium), the sodium salt of Bucladesine, is a cell-permeable PKA activator and a cAMP analog that mimics the action of endogenous cAMP.

ProtocolIn VitroBoth choline acetyltransferase (ChAT) and vesicular acetylcholine transporter (VAChT) mRNA increased approximately fourfold after treatment of PC12 cells with Bucladesine (dibutyryl cyclic AMP; dbcAMP). ChAT and PKA activity are also increased by Bucladesine
In VivoIntrahippocampal infusion of Bucladesine into the CA1 region (male Albino-Wistar rats) can cause an improvement in spatial memory in maze task. Bilateral infusion of 10 μM and 100 μM Bucladesine leads to a significant reduction in escape latency and travel distance (showing an improvement in spatial memory). Bucladesine via activation of PKA and induction of cAMP/PKA pathway improved spatial memory retention

Method for preparing DMSO master liquid: mg drug pre-dissolved in µL DMSO(Master liquid concentration mg/mL) ,Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug.
Method for preparing in vivo formulation: Take µL DMSO master liquid, next add µL PEG300, mix and clarify, next add µL Tween 80,mix and clarify, next add µL ddH2O,mix and clarify.
Note:
  • (1) Please be sure that the solution is clear before the addition of next solvent. Dissolution methods like vortex, ultrasound or warming and heat may be used to aid dissolving.
  • (2) Be sure to add the solvent(s) in order.
