Protocol | In Vitro | Riluzole is an anticonvulsant that belongs to the family of use-dependent Na-channel blockers that also inhibit GABA uptake, IC+50 is 43 μM. At 20 μM, Riluzole only slightly suppresses peak self-touching IPSC, but reliably lengthens IPSC. Riluzole was also found to cause intense, concentration-dependent, reversible enhancement of the 2 μM GABA response. At higher concentrations of Riluzole, especially 300 μM, GABA currents exhibit significant desensitization upon prolonged co-exposure to 2 μM GABA and Riluzole. Riluzole enhances the EC of GABA reactions50 Approximately 60 μM |
